Overview

Apramycin is an aminocyclitol antibiotic (closely related to the aminoglycoside class) primarily used in veterinary medicine for the treatment of enteric bacterial infections in livestock, such as porcine E. coli bacillosis, and in calves and poultry.

While no longer commercially available in the USA, it remains in use internationally. It is notable for its efficacy against a variety of gram-negative organisms. Because it is poorly absorbed from the gastrointestinal tract in adult animals, it is highly effective for local enteric infections. However, neonates may absorb the drug systemically, increasing the risk of classic aminoglycoside toxicities (nephrotoxicity and ototoxicity).

Mechanism of action

Apramycin is bactericidal. It actively transports across the bacterial cell membrane and binds irreversibly to the 30S ribosomal subunit β†’ interferes with mRNA translation β†’ inhibits bacterial protein synthesis.

It is active against many gram-negative bacteria (e.g., E. coli, Pseudomonas, Salmonella, Klebsiella, Proteus, Pasturella, Treponema hyodysenteriae, Bordetella bronchiseptica), as well as Staphylococcus and Mycoplasma.

Storage & stability

Store powder in a cool dry place, in tightly closed containers, protected from moisture. Store at temperatures less than 25Β°C. If exposed to rust, the drug can be inactivated. Prepare fresh water daily. Shelf life of the powder is 24 months.
