概要

ロチラネルはイソキサゾリン系に属する全身性外部寄生虫駆除薬であり、犬および猫のノミ・マダニ感染の治療および予防に使用される。

​臨床のポイント：​ イソキサゾリン系は非常に有効で即効性があるが、有効成分に曝露されるためには寄生虫が動物に付着して吸血を開始する必要がある。

作用機序

Lotilaner acts as an antagonist at gamma-aminobutyric acid (GABA)-gated chloride channels and, to a lesser extent, glutamate-gated chloride channels (GluCls) in insects and acarines.

Binding blocks the transfer of chloride ions across cell membranes → uncontrolled neuromuscular activity → paralysis → death of the parasite.

安全性・警告

禁忌

  • Hypersensitivity to the active substance
  • Puppies less than 8 weeks of age or weighing less than 1.3 kg (dog product)
  • Kittens less than 8 weeks of age or weighing less than 0.5 kg (cat product)

有害事象

  • Vomiting
  • Diarrhea
  • Lethargy
  • Anorexia
  • Neurological signs (tremors, ataxia, seizures) - rare

注意事項

Use with caution in dogs and cats with a history of seizures or neurologic disorders. Must be administered with food or within 30 minutes of feeding for optimal absorption.

Rapidly absorbed when administered with food. Peak plasma concentration is reached within 2 hours in dogs and 4 hours in cats.

Highly protein bound (>99%).

Minimal hepatic metabolism.

消失

Primarily excreted unchanged in feces via biliary excretion; minor renal excretion.

過剰投与

Lotilaner is generally well tolerated even at multiple times the maximum recommended dose. Overdose may cause mild, transient gastrointestinal signs such as vomiting or diarrhea. Treatment is symptomatic and supportive.

薬が適切に吸収されるよう、1か月に1回、​食事と一緒に​または食後30分以内にチュアブル錠を投与してください。この薬はノミやマダニを駆除しますが、薬効を発揮するためには寄生虫がペットを噛む必要があります。
