概要

​メクリジン (Meclizine)​ は第一世代のピペラジン系抗ヒスタミン薬であり、獣医療では主に制吐作用および動揺病（乗り物酔い）の予防に使用されます。

  • ​臨床的有用性​：犬や猫の末梢性前庭疾患、めまい、乗り物酔いの管理に特に有効です。

作用機序

Meclizine acts primarily as an antagonist at central H1-receptors and muscarinic acetylcholine receptors.

  • Vestibular Pathway: Blocks histaminergic and cholinergic signals originating from the vestibular apparatus → reduces stimulation of the ​Chemoreceptor Trigger Zone (CTZ)​ and the vomiting center in the medulla.
  • Anticholinergic Activity: Central muscarinic antagonism contributes significantly to its efficacy against motion sickness and vertigo.
  • CNS Depression: Causes generalized CNS depression, which may aid in its antiemetic and sedative effects.

注意事項

Use with caution in patients with prostatic hypertrophy, bladder neck obstruction, severe cardiac failure, angle-closure glaucoma, or pyeloduodenal obstruction. Because it is an antihistamine, it may affect the results of skin tests using allergen extracts; do not use within 3-7 days before testing. May potentially inhibit lactation due to anticholinergic activity.

過剰投与

Moderate overdosage may result in drowsiness alternating with hyperexcitability.

Massive overdosages may result in:

  • Profound CNS depression
  • Hallucinations
  • Seizures
  • Severe anticholinergic effects (tachycardia, urinary retention, etc.)

Treatment:

  • Consider gut emptying when patients present soon after ingestion.
  • Treatment is symptomatic and supportive.

  • Warning: Avoid respiratory depressant medications.
