概要
オルサラジンナトリウムは、主に獣医学において犬の慢性大腸炎の治療に使用される局所作用型の消化管抗炎症薬です。
サラゾスルファピリジンに反応しない、または耐えられない（多くの場合、サルファ剤部分が乾性角結膜炎などの副作用を引き起こすため）患者の代替薬として機能します。構造的には、アゾ結合によって連結された2分子の5-アミノサリチル酸（5-ASA、メサラジン）からなるプロドラッグです。サラゾスルファピリジンに見られるスルファピリジン部分を持たないため、免疫介在性の副作用を誘発するリスクは一般的に低いですが、涙液産生の問題が時折発生する可能性があります。
作用機序
Olsalazine is a prodrug that passes through the stomach and small intestine largely intact.
Upon reaching the colon, bacterial azoreductase enzymes (produced by normal colonic flora) cleave the azo bond, releasing two molecules of 5-ASA (mesalamine).
- 5-ASA acts locally on the colonic mucosa.
- It inhibits the cyclooxygenase (COX) and lipoxygenase (LOX) pathways → decreasing the production of pro-inflammatory prostaglandins and leukotrienes.
- It also acts as a free radical scavenger, reducing oxidative tissue damage in the inflamed colon.

禁忌
- Hypersensitivity to olsalazine or salicylates
有害事象
- Keratoconjunctivitis sicca (KCS / dry eye) - less frequent than with sulfasalazine
- Diarrhea (paradoxical worsening reported in humans)
- Vomiting (especially with overdose)
- Decreased motor activity (with overdose)
- Elevated ALT or AST
注意事項
Important Note: While the risk of Keratoconjunctivitis Sicca (KCS) is lower than with sulfasalazine, baseline and periodic Schirmer Tear Tests (STT) are still highly recommended.
- Renal Disease: Use with caution in animals with pre-existing renal disease, as rare nephrotoxicity has been reported in human patients.
- Pregnancy (FDA Category C): High-dose rat studies showed some fetal abnormalities. Use during pregnancy only when benefits outweigh the risks.
- Lactation: Oral olsalazine given to lactating rats produced growth retardation in pups. Use with caution in nursing patients.
- Hepatic: May cause increases in ALT or AST; monitor liver enzymes if used long-term.

Poorly absorbed systemically; approximately 98% of an oral dose reaches the colon intact.

Acts locally within the lumen and mucosa of the colon.

Cleaved in the large intestine by bacterial azoreductases into 5-aminosalicylic acid (5-ASA, mesalamine).
消失
The small amount of drug that is absorbed systemically is rapidly eliminated.
過剰投与
Overdosage in dogs may cause vomiting, diarrhea, and decreased motor activity.
- Toxicity: Dosages up to 2 grams/kg were not lethal in dogs.
- Treatment: Treat symptomatically and supportively. GI decontamination may be considered if a massive overdose was recently ingested.

保管・安定性
Store capsules at room temperature. Olsalazine sodium is stable under physiologic acidic and alkaline conditions.
飼主向け情報
- 投与方法： 胃の不調を最小限に抑えるため、食事と一緒に与えてください。1日の投与間隔はできるだけ均等にしてください。
- ドライアイの観察： ドロッとした黄緑色の目やに、目を細める、または目の充血に注意してください。この薬は時折、涙の分泌を減少させる（「ドライアイ」またはKCSと呼ばれる状態）ことがあります。
- 便の観察： この薬の投与を開始した後、犬の下痢が悪化した場合は獣医師に連絡してください。
獣医師の指示がない限り急に投与を中止しないでください。副作用が現れた場合は相談してください。
