概要

​プロリゲストン​は、主に犬や猫の発情の延期または抑制に使用される合成プロゲストーゲンです。猫の粟粒性皮膚炎の治療にも認可されていますが、この適応症には通常、特定のグルココルチコイドが推奨されます。

​臨床上の警告​: プロリゲストンには、嚢胞性子宮内膜増殖症（子宮蓄膿症）、糖尿病、乳腺腫瘍の発生リスクなど、長期的なプロゲストーゲン使用に特有のリスクがあります。慎重に使用する必要があり、糖尿病の動物には禁忌です。

作用機序

Proligestone alters the transcription of DNA → leads to alterations in cellular metabolism → mimics the physiological effects caused by endogenous progesterone.

安全性・警告

禁忌

  • Diabetic animals (insulin requirements are likely to change unpredictably)
  • Bitches before or at their first oestrus

  • Unpredictable duration of anoestrus (up to 3 years in some bitches)
  • Temperament changes (listlessness and depression)
  • Polyuria and polydipsia (increased thirst)
  • Polyphagia (increased appetite)
  • Cystic endometrial hyperplasia / Pyometra
  • Diabetes mellitus
  • Acromegaly
  • Adrenocortical suppression
  • Mammary enlargement, lactation, or neoplasia
  • Injection site irritation
  • Coat colour changes at the injection site
  • Calcinosis circumscripta at the injection site

注意事項

As coat colour changes may occasionally occur, injection into the medial side of the flank fold is recommended for thin-skinned or show animals.

Clinical Pearl: While proligestone does not appear to be associated with as many or as serious adverse effects as older progestogens (e.g., megestrol acetate, medroxyprogesterone acetate), owners must still be thoroughly counseled on the risks of pyometra and diabetes mellitus.
