개요
클로르술론(Clorsulon)은 주로 흡충구충제로 사용되는 매우 효과적이고 좁은 스펙트럼의 항기생충제입니다.
- 주요 적응증: 소의 미성숙 및 성충 간흡충(Fasciola hepatica) 감염 치료제로 FDA 승인을 받았습니다.
- 허가 외 사용: 양과 낙타과(라마 등)의 흡충 감염에도 자주 사용됩니다.
- 제한 사항: 8주령 미만의 미성숙 흡충 및 반추위 흡충(Paramphistomum spp.)에는 효과가 없으며, 양의 거대간흡충(Fasciola magna)을 완전히 박멸하지는 못합니다.
임상 요점: 스펙트럼이 좁기 때문에 클로르술론은 종종 매크로라이드계 약물(예: 이버멕틴)과 혼합되어 선충과 흡충 모두에 대한 광범위한 구충 효과를 제공합니다.
작용 기전
Clorsulon exerts its flukicidal effect by disrupting the parasite's energy metabolism.
- It competitively inhibits two key glycolytic enzymes in susceptible flukes: 3-phosphoglycerate kinase and phosphoglyceromutase.
- This blockade disrupts the Emden-Myerhof glycolytic pathway → depletes the fluke's primary metabolic energy source (ATP) → leads to paralysis and death of the parasite.
- Dose-dependent efficacy: At 7 mg/kg, it kills migrating flukes (8 weeks post-infection); at 2 mg/kg, it only affects adult flukes (14 weeks post-infection).

주의
- Dairy Cattle: No milk withdrawal time has been determined; labeled not for use in female dairy cattle of breeding age.
- Administration: The combination injectable product (Ivomec Plus®) must be administered subcutaneously only; do not give IV or IM.
- Species Warning: The manufacturer warns to use the combination product in cattle only, as severe reactions, including fatalities in dogs, may occur.

After oral administration to cattle, the drug is absorbed rapidly with peak levels occurring in about 4 hours.
분포
Approximately 75% of the circulating drug is found in plasma and 25% in erythrocytes. At 8-12 hours after administration, clorsulon levels peak in the fluke.
과다투여
Clorsulon is very safe when administered orally to cattle or sheep. Doses of up to 400 mg/kg have not produced toxicity in sheep. A dose that is toxic in cattle has also not been determined.
