概覽
更昔洛韋是一種抗病毒藥物，主要用於治療貓皰疹病毒-1型 (FHV-1) 的眼部感染。體外研究顯示，其對抗FHV-1的效果至少是阿昔洛韋 (aciclovir) 的10倍。
臨床警告： 本藥無法根除潛伏的病毒感染。雖然臨床軼事報告顯示其局部使用效果良好，但尚未有針對貓的完整安全性或藥代動力學報告。在難治性和嚴重的FHV-1潰瘍病例中，可採用包含局部或全身性抗病毒藥物的聯合療法。
作用機制
Ganciclovir inhibits viral replication by targeting viral DNA polymerase. It depends on viral thymidine kinase for initial phosphorylation → converted to active ganciclovir triphosphate by cellular enzymes → competitive inhibition of viral DNA polymerase → termination of viral DNA chain elongation.
